Abstract/Contents: | "Although Kentucky bluegrass (Poa pratensis L.) cultivars have been developed with good turfgrass quality, irrigation is often necessary to maintain quality during heat and drought stress periods. Inter-specific hybridization between Texas bluegrass (Poa arachnifera Torr.), a native to Texas and Oklahoma, and Kentucky bluegrass is currently being utilized to incorporate genes for heat and drought tolerance into Kentucky bluegrass. A test to determine true hybrids would be useful to breeding programs. The objective of this study was to develop SCAR (Sequence Characterized Amplified Region) markers specific for Kentucky bluegrass and Texas bluegrass and use them to identify F1 and BC generation hybrids between these two species. SCAR markers were developed from sequencing a single RAPD (Random Amplified Polymorphic DNA) band and designing primers to amplify the chosen band in Kentucky or Texas bluegrass. Eight Kentucky bluegrass cultivars, four female Texas bluegrass plants and 12 hybrids from F1 through BC3 generations were analyzed with the SCAR primer pairs. The respective SCAR markers differentiated between Kentucky bluegrass and Texas bluegrass plants in all cases. All F1 and BC1 hybrids amplified both Kentucky and Texas bluegrass SCAR markers. The Texas bluegrass SCAR marker did not amplify in some BC2 and the BC3 hybrid. These data indicate that these hybrids presumably lost the Texas bluegrass marker due to introgression of the Kentucky bluegrass DNA where the SCAR marker exists. The species-specific SCAR markers developed in this study will be useful to breeding programs utilizing inter-specific hybridization between Texas and Kentucky bluegrass." |
